No you don't need to already level it to 50.
My templar is using assassination and my highest level nightblade is around level 30 and hasn't maxed out any of the skill lines yet. I also had the option of choosing arcanist skills and I've never even made an arcanist. (I should, I've been planning one for ages, but haven't gotten around to it yet.)

I leveled to 50 all my class lines on each class during jubilee because I understood it was needed to be able to access them with subclassing... If so, it was just a waste of time then...
Lots of confusion and conflicting informations around the system.

Yeah, I felt the same. Everyone thinks something different so it's good to get info from players who've already done it.
