I mostly say "E SO" although I do find myself saying "E S O" sometimes, I think it depends who I am talking with.
Edit: To mention as well I am from the UK, as I wonder if there is a region based pattern on how people say it.
When pronouncing an Abbreviation, you spell out each letter individually, unless the abbreviation was purposely made to create an Acronym, of which then you would pronounce the word the Abbreviation makes. That is, i believe, the proper grammar.
ESO is not meant to create an actual word, so thus i say it as E, S, O.

And so I treat it the same way.
Plus, every time I have watched them, ZOS staff, from Matt Firor down, treat it as such.
