Yeah indeed. The way I usually do it, so as to not affect in the settings which ones I actually want on or off, is to log off, find the addons download folder, and rename it to "AddOns-off", then start up the game again and log back in. When I want them back on again, I go back to the folder and rename it back to "AddOns" (or, if the game has recreated the AddOns folder, I just move them into that one). Otherwise, as Scipio said, you can only have one "Kill Enemy <x>" quest available at a time, so you would need to complete or abandon any current ones before you can pick up a new one.
Hmm, not sure what else to suggest. Can you upload a couple of screenshots of your map, your journal, and your character standing by the mission boards?
