I would probably guess on number 2. Not sure why but just a feeling :P

With all your characters being Redguards, I don't think resource management is an issue so I am not sure 5-piece Vicious Serpent is a good choice in Trials. I would lean toward the second setup with TBS + Alkosh. But why not 5-piece Alkosh? Is someone else in group already running it? Seems like somebody should be running 5 Alkosh and you are only 1 piece away in that setup. I am a big fan of group-wide armor debuffs like 5 NMG, 5 Sunderflame, 5 Alkosh, etc.

The difference between 1 and 2 is a weapon critical bonus vs. a weapon damage bonus, right? I believe the critical is better, and all other serpent's buffs are just nice extras.

Setup 3 gives too little health, while setup 4 is an easy-to-get setup for new players, I think, rather than a real endgame proposal.

The setup 4 is a "CBA farming SO" one to be honest...

Crit multipliers probably too good to let TBS pass on with setup 3. Plus no health bonus, so...

I would actually make a setup 2 bis and replace the 4th Alkosh by the 2nd Kena. Even a 30% uptime will outperform a single weapon damage bonus, but it depends on rotation (can trigger it during Hail refresh / swap). If not, setup 2 is my pick among those 4.

Setup 1 is second best, by a very small margin.

Is setup 4 good for trials and VMA? Would love to know as resources are scarce. Ty

All setups assume that someone else in the group is applying 5pc NightMother's Gaze debuff to the target, and a second one would be useless.

Imo, set 1 for trash and swap out helm/shoulder for 1pc molag/veli for boss.

Too bad I didn't get my medium veli divine shoulder after 150 keys.

By specs, and percentages for certain. I feel more-better-er with my TBS on though.

Night Mother's Gaze increases DPS more than TBS and in fact more than anything else. But as it is a debuff, it requires only one person in a group to use it. So if someone in your group is already using it, you are already benefiting from it and other sets would help you more.
