If you already have a Legendary (Gold) item, adding a Legendary Rune to it won't make any difference, right?

The enchant on a dropped gold item is already gold quality, yes.
The enchant on a dropped item of less than gold quality, when the item is improved to gold quality also improves the enchant to gold quality.
UNLESS you have changed the enchant yourself prior to improving the item, in which case the player-added enchant remains at its prior quality.
