top atm is vo/alk + sprig/tbs + kraugh/veli/selene + ma
you should start with 5 hund, 5 sprig, 2 monster/agil and build up from there

I actually tested Armor Values in vMA and in most cases, both Spriggan and TFS are overdoing it:
As you need to stack Penetration with TFS, its extra bad for vMA where burst is far more benefical as dps. Most targets die in 1-3 hits so you will have a hard time to keep the stacks up.
The issue with Twice Fanged Serpent is that the Penetration is just too much. There are very few occations where you can actually benefit from 5k Pen.
You'll get 5280 from Major Fracture and 5160 from Sharpened Weapons = 10.440 Penetration. Overland Mobs have 9.1k, which means for questing, you are already overdoing it.
In Dungeons/Trials, we face 18.2k Armor targets.
Adding Kra'gh which is considered BiS atm, we get another 1935 Penetration which brings us to 12.375.
In a group, a tank or a healer usually uses a Crushing Glyph, another 1622 Penetration -> 13.997
Now if we add 5k Pen from TFS, we're already at 18997, overdoing it by 797 already.
And we didnt even talk about Alkosh or Minor Fracture yet.
So there are very limited uses of TFS in PvE.

Isn't Kra'gh is only BiS because of the penetration? If you use TFS, you don't need Kra'gh and you can use a much stronger set, like Stormfist or Velidreth. Stam builds are lacking AoE damage. Both Stormfist and Velidreth have much more AoE potential than Kra'gh. You can also use a Precise weapon if you have a decent uptime on Alkosh.
I don't know about Minor Fracture, stamina templars are rare...
TFS only does what light armor have as a passive. So if magicka builds aren't over penetrating, then how would a stamina build over penetrate?
