Do staves count as two-handed weapons?

    No. They count as either destruction or restoration, depending on what type they are.

    They do count as a two handed weapon, however they will not give you access to the two handed melee skill line, only the two handed ranged magical skill lines, which are destruction/restoration.

    It takes an entirely different skill set to point a stick at someone than it does to chop them in half with a really big axe. ;)

    Weapon passives only ever work if you have that weapon active (having it on your weapon swap doesn't count). So passives from the Two-Handed skill line state they only work when you are using a two-handed melee weapon.
