If I already have one of each class maxed out on all skill lines, do I need to level those skill lines again if I pick any to sub in?

Yes, you need to level subclass skill lines. For eg., if you are on an arcanist and you want to get the necro gravelord skill line, you will have to level the skills from scratch, even if you have a maxed out necro character, afaik. The subclassed skills cost 2 skill point instead of 1, and they level slower than native class skills. On the upside, if you level a subclass skill line on one character, it will be unlocked for all your other characters, so from the previous example, if you level the gravelord skill line on an arcanist, and you also want to have it on your NB, you don't have to level it again on your NB, it will be already available.
